Manuel Parker, known as Spider-Man was killed in his home world by the Sinister Six, led by the Green Goblin. Luna, Celestia, Twilight and Spike teleported to NYC and found out that Spidey did die, but brought him back to life and teleported him to Equestria for a second chance. He would see some familiar faces in Starlight Glimmer and Sunset Shimmer and reunites with them as well.

You are going to see some of the rivals from Spidey's rogue gallery, as Manuel Parker (Spider-Man) would try to survive in the world of KFP, while being hunted down by his most hated rival, Norman Osborn (the Green Goblin).
